چکیده

Abstract The Betula genus contains pentacyclic triterpenoid betulin known for its environmental adaptation and medicinal properties. However, the mechanisms underlying biosynthesis responding to climate change remain unclear. In this study, role of epigenetic modification (DNA methylation) in was examined how climatic factors influence it. Whole-genome bisulfite sequencing performed greenhouse-grown Chinese white birch ( platyphylla Sukaczev) treated with DNA methylation inhibitor zebularine (ZEB) a natural population Northeast China. ZEB treatment significantly affected CHH level transposable elements content hormesis dose-dependent manner. expression bHLH9, key transcriptional factor controlling biosynthesis, were also consistently by as hormetic dose–response. population, there positive correlation between promoter bHLH9 summer precipitation, while winter temperature negatively correlated. Thus climate-dependent regulates downstream genes involved biosynthesis. This study highlights signals induce changes that result production, possibly helping develop resilient plants combat ongoing enhance secondary metabolite production.

Epigenetic Modification of the Repair Donor Regulates Targeted Gene Correction

Optimizing design of vectors is critical to effective gene therapy. In targeted gene correction (TGC), cleavage of chromosomal DNA near a mutation stimulates homology-directed repair of a target gene using a donor provided in trans. We have systematically addressed epigenetic parameters of donor design, using a flow-based assay to quantify correction frequencies and expression levels of a green...

Study of the betulin enriched birch bark extracts effects on human carcinoma cells and ear inflammation

UNLABELLED BACKGROUND Pentacyclic triterpenes, mainly betulin and betulinic acid, are valuable anticancer agents found in the bark of birch tree. This study evaluates birch bark extracts for the active principles composition. RESULTS New improved extraction methods were applied on the bark of Betula pendula in order to reach the maximum content in active principles. Extracts were analyzed ...

Epigenetic Modification of TLRs in Leukocytes Is Associated with Increased Susceptibility to Salmonella enteritidis in Chickens

Toll-like receptors (TLRs) signaling pathways are the first lines in defense against Salmonella enteritidis (S. enteritidis) infection but the molecular mechanism underlying susceptibility to S. enteritidis infection in chicken remains unclear. SPF chickens injected with S. enteritidis were partitioned into two groups, one consisted of those from Salmonella-susceptible chickens (died within 5 d...

Acetylation of H2A.Z is a key epigenetic modification associated with gene deregulation and epigenetic remodeling in cancer.

Histone H2A.Z (H2A.Z) is an evolutionarily conserved H2A variant implicated in the regulation of gene expression; however, its role in transcriptional deregulation in cancer remains poorly understood. Using genome-wide studies, we investigated the role of promoter-associated H2A.Z and acetylated H2A.Z (acH2A.Z) in gene deregulation and its relationship with DNA methylation and H3K27me3 in prost...

Epigenetic modification of the human CCR6 gene is associated with stable CCR6 expression in T cells.

CCR6 is a chemokine receptor expressed on Th17 cells and regulatory T cells that is induced by T-cell priming with certain cytokines, but how its expression and stability are regulated at the molecular level is largely unknown. Here, we identified and characterized a noncoding region of the human CCR6 locus that displayed unmethylated CpG motifs (differentially methylated region [DMR]) selectiv...
